CHEF1 Expression System

CMC’s proprietary cell line expression system, CHEF1®, is available to our clients. The CHEF1 system results in substantially higher expression levels than those obtained with other vectors.
CHEF1 requires no gene amplification step, thus saving many months in cell line development. CHEF1 also permits co-expression of genes encoding other important factors or subunits.

Further, the availability of a large library of vectors possessing different selectable markers permits transfection of cell lines with vectors encoding additional copies of the same gene, thereby increasing expression levels to still higher levels. CHEF1 also permits co-expression of genes encoding other important factors or subunits.

In comparison with conventional vectors, the CHEF1 vectors provide:

  • Expression levels, without amplification, that exceed by 6- to 25-fold those obtained with other promoter/vector systems such as hCMV.
  • Clone stability in the absence of methotrexate.
  • A variety of selectable markers that permit high-level co-expression of protein subunits.
    For example, by transfection with multiple CHEF1 plasmids -- carrying the heavy chain, light chain or both heavy and light chain genes -- CMC Biologics has been able to obtain high-level and balanced expression of both antibody subunits.
